The Los Angeles awards show, organised by officials at the National Association for the Advancement of Colored People (NAACP), saw big wins for Denzel Washington, who was named Outstanding Actor for The Book of Eli, and Halle Berry, Outstanding Actress for Frankie & Alice. Another big star, Samuel L. Jackson, received the Outstanding Supporting actor prize for Mother and Child.
But it was Perry's film, starring Whoopi Goldberg and Janet Jackson, which took the top honours, winning Best Film of 2010 as well as the best supporting actress trophy for Kimberly Elise.
The actor/writer/director said of his win, "I wanted to take this iconic work and introduce it to people who had never heard of it... There is nothing like having nine strong black women holding you up."
Perry also triumphed in the TV categories, with Tyler Perry's House of Payne named Outstanding Comedy Series, and the show's star, David Mann, earning the award for Outstanding Actor in a comedy.
Other prizes went to Vanessa Williams, who was named Outstanding Actress in a Comedy Series for her turn in Desperate Housewives, and Sofia Vergara was honoured for her performance in Modern Family, while Grey's Anatomy took the Outstanding Drama Series prize.
The winners also included Ice Cube, LL Cool J, Regina King, Terrence Howard, Idris Elba and Will Smith's 10-year-old daughter Willow, who won the best new music artist award ahead of Bruno Mars and rapper Nicki Minaj.
